TUDCA (Tauroursodeoxycholic Acid)

Mechanism of Action

TUDCA is a water-soluble bile acid conjugate (taurine + ursodeoxycholic acid) with potent hepatoprotective and cytoprotective properties. It inhibits the mitochondrial pathway of apoptosis by preventing BAX translocation to mitochondria and cytochrome c release. TUDCA reduces endoplasmic reticulum (ER) stress by acting as a chemical chaperone, assisting protein folding and reducing the unfolded protein response (UPR). It protects hepatocytes by stabilizing cell membranes, displacing toxic hydrophobic bile acids (chenodeoxycholic, lithocholic) from biliary epithelium, and promoting choleresis (bile flow). Additionally, it has neuroprotective effects via reduction of ER stress in neurons and modulation of the TGR5 bile acid receptor.

Dosing Protocol

Standard: 250-500mg daily

Loading: 1000-1500mg/day for acute liver support or hepatotoxic drug exposure (short-term, under supervision)

Maintenance: 250mg/day

Administration: oral

Timing: With meals for liver support. Some protocols recommend before meals to prime bile flow.

Duration: Cycle 4-8 weeks for liver support; ongoing at low dose for chronic liver conditions under supervision

Notes

TUDCA is the premium liver support compound — more potent and better absorbed than UDCA (ursodeoxycholic acid, which is the prescription form used in primary biliary cholangitis). UDCA requires bacterial deconjugation and reconjugation, while TUDCA is already in the active, taurine-conjugated form. The ER stress reduction mechanism extends its potential beyond liver disease to neurodegenerative conditions (ALS, Parkinson's, Huntington's — active clinical trials). The bodybuilding community uses TUDCA extensively for liver support during oral anabolic steroid cycles — while we don't endorse that application, the hepatoprotective evidence supporting TUDCA in toxic exposure is real. Interactions

  • Other bile acid sequestrants (cholestyramine) [MEDIUM] — Bile acid sequestrants bind TUDCA in the gut, reducing its absorption
  • Aluminum-containing antacids [LOW] — May reduce TUDCA absorption
  • Cyclosporine [LOW] — TUDCA may alter cyclosporine absorption via bile flow modulation
  • May reduce absorption of other medications (bile acid sequestrant effect)
  • Synergistic with NAC for liver protection

Contraindications

  • Complete biliary obstruction (cannot reach bile ducts if obstruction is complete)
  • Calcified gallstones (UDCA/TUDCA dissolve cholesterol stones, not calcified)
  • Acute cholangitis
