Nootropics
Evidence: animal_plus_anecdotal
Seed oil contains sesquiterpenes, alkaloids (celastrine, paniculatin), and polyunsaturated fatty acids that demonstrate dose-dependent cholinergic activity by inhibiting acetylcholinesterase (AChE) and increasing acetylcholine levels in hippocampal and cortical regions. Provides neuroprotection against oxidative stress through elevation of glutathione (GSH), catalase (CAT), and superoxide dismutase (SOD). Seed extracts attenuate hydrogen peroxide- and glutamate-induced excitotoxic injury in neuronal cells, and exhibit anti-inflammatory and anxiolytic properties.
Standard: 500-1000 mg/day of seed extract (standardized to >8% polyphenols) or 10-15 seeds daily (traditional Ayurvedic dosing, escalated from 1 seed/day)
Maintenance: 500 mg/day of standardized extract
Administration: oral
Timing: Morning, with or without food. Traditional practice involves gradual dose escalation starting from 1 seed daily, increasing by 1 seed per day up to 10-15.
Duration: Traditionally used in 30-60 day courses. No established cycling protocol from clinical data.
Known as the 'Intellect Tree' in Ayurvedic medicine (Jyotishmati). The traditional gradual dose escalation (1 seed/day, increasing by 1 seed daily) is designed to build tolerance and may have pharmacological rationale. Evidence base is almost entirely animal studies and traditional use — no published RCTs in humans for cognitive outcomes as of 2025. The seed oil has been shown to cross the BBB and accumulate in brain tissue. Promising preclinical data for both neuroprotection and cognition, but human validation is lacking. A reasonable adjunct to a nootropic stack based on traditional evidence, but should not be relied upon as a primary cognitive enhancer.
Primarily available as seed oil or seed extract from Ayurvedic/nootropic vendors. Standardization is inconsistent — polyphenol content is the most common marker (look for >8%). Nootropics Depot offers a SmartSeed extract standardized to polyphenol content. Raw seeds are available but dosing precision is poor. No pharmaceutical-grade product exists.
