Active triterpenoid saponins (bacosides A and B) provide multifaceted neuroprotection: inhibition of acetylcholinesterase (AChE) and activation of choline acetyltransferase (ChAT) to enhance cholinergic neurotransmission; upregulation of tryptophan hydroxylase and serotonin transporter expression to modulate serotonergic tone; antioxidant neuroprotection via induction of superoxide dismutase and glutathione peroxidase; and reduction of beta-amyloid aggregation. Additionally enhances GABA signaling through GABA-A receptor subunit upregulation and glutamate decarboxylase activation.
Standard: 300-450 mg/day of standardized extract (24-55% bacosides, typically Bacognize or Synapsa brands)

Timing: With a fat-containing meal to improve absorption of fat-soluble bacosides. Morning or evening — some users report mild sedation. Minimum 8-12 weeks for full cognitive effects.

One of the most rigorously studied natural nootropics, with a 2014 meta-analysis of 9 RCTs confirming improvements in attention speed. The GI side effects are the primary limiting factor — taking with food and starting at a lower dose mitigates this. Effects take 8-12 weeks to manifest; many users abandon it too early. The anxiolytic/adaptogenic effects often appear before cognitive enhancement. May mildly increase thyroid hormones — relevant for those on thyroid medication. Historically used in Ayurveda as 'Brahmi' for memory enhancement in children.
Standardized extracts are essential — raw herb is inconsistent. Synapsa (CDRI 08, 55% bacosides by HPLC) and Bacognize (45% bacosides, serotonin-standardized) are the two most clinically validated brands. Generic 'bacopa extract' products vary wildly in bacoside content. Fat-soluble — capsule or powder with fat preferred.
