Active triterpenoid saponins (bacosides A and B) provide multifaceted neuroprotection: inhibition of acetylcholinesterase (AChE) and activation of choline acetyltransferase (ChAT) to enhance cholinergic neurotransmission; upregulation of tryptophan hydroxylase and serotonin transporter expression to modulate serotonergic tone; antioxidant neuroprotection via induction of superoxide dismutase and glutathione peroxidase; and reduction of beta-amyloid aggregation. Rhodiola's key bioactives are rosavin (unique to R. rosea), salidroside, and tyrosol. It modulates the HPA axis stress response by inhibiting cortisol release and stress-activated protein kinases (SAPKs/JNK). Salidroside activates AMPK, enhances mitochondrial biogenesis via PGC-1alpha, and stimulates glucose uptake. Rhodiola inhibits monoamine oxidase A and B (MAO-A, MAO-B), increasing synaptic serotonin, dopamine, and norepinephrine levels. It also stimulates Nrf2 antioxidant defense, increases erythropoietin (EPO) production, and enhances beta-endorphin levels during exercise.
