Vitamin D3 (cholecalciferol) is hydroxylated in the liver to 25(OH)D (calcidiol), then in the kidneys to 1,25(OH)2D (calcitriol), the active hormone. Calcitriol binds the nuclear vitamin D receptor (VDR), forming a heterodimer with RXR that regulates >1000 genes. Key targets: osteocalcin (directs calcium into bone matrix), matrix Gla protein (MGP, inhibits arterial calcification), Gas6 (cell signaling, neuroprotection), and protein S (anticoagulant). MK-7 has a long half-life (~72 hours vs 1-2 hours for K1) enabling consistent carboxylation activity with once-daily dosing.
